研究簡介:
      急性缺血中風(ischemic stroke)是全球先進國家高死亡率的疾病。中風的病理機制十分複雜,其中以養份(葡萄糖跟氧氣)缺乏(oxygen andglucose deficiency; OGD)所造成的興奮性胺基酸毒(excitotoxicity)、粒線體損傷、氧化/硝化壓力(oxidative/nitrosative stresses)、發炎(inflammation)及細胞壞死/凋亡(necrosis/apoptosis)等機制最為重要。我們發現中藥複方(B7及T11)可以顯著延長缺血型中風(ischemic stroke)小鼠的存活時間,比正對照組藥物rt-PA(血栓溶解劑)及MK-801(NMDA受體拮抗劑)來得更有效。中風鼠經MK-801及rt-PA治療可延長部份動物存活至第五天(未治療中風鼠3天內全數死亡),但投予B7T11則可以使4-6成動物存活率達七天以上,並有效減少動物大腦皮質(cortex)及海馬迴(hippocampus)之神經元壞死,改善神經功能障礙(neurological deficits),及促進神經再生。基於B7方及T11對缺血型中風之治療效果卓著,又臨床上目前並無有效治療藥物,本研究室擬進一步追蹤分離B7T11活性部分純化物及其中所含的活性化合物,並進行分子機制探討,最終擬將其發展成為治療缺血型中風相關的藥物。
