藥用石斛新品種-種苗金皇一號石斛

摘要

金皇石斛在預防與緩解乾眼症狀之食品開發

新雜交育成藥用石斛新品種金皇石斛(Dendrobium Taiseed Tosnobile;DTT)是由黃花石斛 ( Dendrobium tosaense )金釵石斛( Dendrobium nobile )雜交選育而得,其優勢在於取得黃花石斛生長快速的特性並掌握金釵石斛所含有的有效成分,並已取得英國皇家園藝學會給與之學名及中華民國品種權。先前研究已通過小鼠急慢性毒性分析,並且已發表2SCI報告。由於古籍石斛護眼之記載,但是缺乏實驗證據,因此以小鼠紫外線B誘發乾眼研究模式,探討金皇石斛萃取物預防與緩解乾眼症狀之效果。將6周大之ICR雌鼠隨機分為6組,包含: 對照組、紫外線B照射乾眼組、3個實驗組給予紫外線B並分別餵食金皇石斛樣品各2050100 mg/Kg、以及單純餵食金皇石斛樣品100 mg/Kg組。紫外線B照射及餵食金皇石斛萃取物均每日進行,全部組別於第047天測量淚液量(tear volume; TV)及淚液膜破裂時間(tear film break up time; TBUT),並於第8天染色拍照評估眼球表面變化,隨後犧牲進行病理切片分析。結果顯示眼球表面及淚液分泌量及淚液皆隨著餵食金皇石斛萃取物而得到改善,並具有劑量相關性。病理切片分析結果顯示角膜上皮以及外淚腺及麥氏腺的損傷及發炎狀況得到改善。實驗結果證實金皇石斛萃取物確實有預防與緩解乾眼症狀之功效,可望在其原有生產的優勢上,往前進一步開發緩解乾眼症狀之食品。

金皇石斛在緩解癌症惡病質之食品開發

動物實驗得知金皇石斛水萃物 (75 mg/kg) 及酒精萃物 (150 mg/kg) 皆能減緩癌症惡病質小鼠中體重減輕與肌肉耗損萎縮的症狀並能增加其肌肉握力、飲食取水量與存活率。在細胞實驗中則以肌肉細胞為模式,證明金皇石斛水萃物與酒萃物亦皆可以有效地恢復因博來黴素處理而造成之肌小管形成被抑制的現象,而以水萃物效果最好,而單獨石斛水萃物即能誘發肌纖維母細胞分化成肌小管。顯示石斛水萃物具有與肌肉保護及分化相關的關鍵化合物。

Dendrobium Taiseed Tosnobile as a food ingredient for prevention and alleviation of dry eye symptoms

A new hybrid, Dendrobium Taiseed Tosnobile(DTT), was derived from crossing and breeding between Dendrobium tosaense and Dendrobium nobile. The new hybrid has advantages of speedy growth while maintaining high concentration of effective ingredients. It has the scientific name given by the Royal Horticultural Society and the rights of the Republic of China. Previous studies have proven its safety for diet, as it passed acute and chronic toxicity tests, including 28-day and 90-day feeding toxicity, genotoxicity, and reproductive toxicity, and from which two SCI-indexed reports have been published. Since ancient Chinese medicine had described Dendrobium for vision protection, but without experimental evidence, the effects of Dendrobium Taiseed Tosnobile extracts for alleviation of dry eye symptoms in an UVB-induced mouse model were investigated. Female ICR mice aged 6 weeks were randomly divided into 6 groups: vehicle control group, UVB-induced dry eye group, 3 experimental groups undertaken UVB irradiation and given extracts at 2050100 mg/Kg of body weight, and a group given 100 mg/Kg of body weight without UVB irradiation. The dietary feeding and UVB irradiation were given daily. One days 0, 4, 7, tear volume (TV) and tear film break up time (TBUT) were assessed and photography was taken on day 8 for evaluation of ocular surface damages, before the animals were sacrificed for histopathological analyses. The results showed that the status of ocular surface, TV and TBUT were dose-dependently improved. Histopathological analysis showed amelioration of damages to extra-lacrimal glands and meibomian glands. These results support and justify further development of Dendrobium Taiseed Tosnobile dietary products for dry eye relief. Furthermore, our results showed that both the water and ethanol extracts of DTT effectively reversed the muscle wasting, the weight loss while increased the grip strength, the intake of food and water and survival rate of cancer-cachexia mice. In the in vitro study, addition of water extracts of DTT effectively protected the muscle cells fiber reduced by bleomycin. Furthermore, the treatment of DTT water extracts alone significantly promoted the differentiation of myoblast cells into muscle cells fiber, one or more indicated compounds are existed in the water extracts of DTT.
