研究簡介:
台灣目前已符合世界衛生組織所定義之老化社會。如何運用中醫藥知識促進“健康老化”以提升老年人口生活品質,並延伸研究成果至新藥生技產業的推廣,是我的研究團隊自我期許以及使命。根據研究專長,研究主題鎖定在代謝疾病及與之相關之神經退化性疾病(阿茲海默氏症),結合中藥及天然物研究開發及穴位埋線針灸之療效轉譯等的基礎、臨床轉譯及應用研究,達到運用中醫藥輔助或替代治療老年相關之慢性病。
(1) 藥物活性篩選平臺及療效評估動物模式簡介 (提供合作及委託研究)

細胞平臺
胰島細胞: BRIN-BD11 cell line, 用於提升葡萄醣刺激胰島素分泌活性分析。
肝臟細胞: H4IIE cell line, 用於抑制糖質新生及脂肪肝活性分析。
骨骼肌細胞: Differentiated L6 myotube, 用於葡萄醣吸收活性分析。
脂肪細胞: Differentiated 3T3-L1 adipocytes, 用於抑制脂肪細胞生長及脂質代謝。

動物模式
胰島素依賴型: STZ 誘導之胰島素依賴型糖尿病鼠。
飲食誘導肥胖(DIO): 高脂飲食誘導肥胖鼠。
誘導式第二型糖尿病: 結合高脂飲食及單劑量STZ注射誘導之動物模式(HFSTZ)。
第二型糖尿病: 瘦體素(leptin)受體突變造成自發性第二型糖尿病(db/db mice)。
共病模式: APPswe/PS1de9基因轉殖鼠結合HFSTZ誘導之”代謝異常加速之阿茲海默氏症”小鼠模式。

(2) 尋找中藥及天然物中可調控腸泌素之小分子藥物

腸泌素效應是比較口服葡萄醣所刺激的胰島素是靜脈注射葡萄糖所刺激胰島素分泌總量的四倍而發現的。目前已知有兩種腸泌素: 昇糖激素類胜肽-1 (glucagon-like  peptide-1, 簡稱GLP-1) 以及胃抑素 (gastric inhibitory peptide 或 glucose-dependent insulinotropic polypeptide, 簡稱 GIP)。由於這類胜肽具有促進胰島功能及食慾控制的特性,兩者皆被用來開發成治療糖尿病的胜肽藥物。
我們目前致力於從中藥或天然物成分中開發具口服性之昇糖激素類胜肽-1受體 (簡稱: GLP-1R)活化小分子。現在已有一專利成分具有GLP-1R受體活化及顯著改善第二型糖尿病小鼠的藥理活性。

(3) 穴位埋線針灸(the Acupoint Catgut-Embedding Therapy)之轉譯研究
穴位埋線針灸運用於疾病以及減重的治療已經行之多年。目前我們研究團隊正與臺北市立聯合醫院林森中醫昆明院區(中醫中心)的許中華院長、葉裕祥醫務長、陳怡如醫師等人進行轉譯研究之合作,利用動物模式進行穴位埋線針灸治療及分子機轉之探討。
